The human p53 gene is located at chromosome 17p13.1 and it encodes a 53 kDanuclear phosphoprotein. p53 is the most striking tumor suppressor gene because it ismutated in most types of human cancer arising from a wide spectrum of tissues.p53 genetic mutation and it's expression often followed the product tumorigenesis,andthe patients which express p53 had the poor prognostic parameters,so to detect the geneticmutation made for tumorous diacrisis. p53 genetic mutation lead to the protein propertychanged,and the stability consumedly increased,it could be analyzed by molecular biology,immunohistochemistry, serology. The immunohistochemistry is the most important method todetect the tumors.In this paper ,The prokaryotic expression vector PQE-p53 we used was constructed byour laboratory to found a method to prepare depurated protein and its antibody. Thehybridoma technology is used to prepare anti-p53 monoclonal antibody, which canspecifically bind to human p53 protein. And we have raised one hybridoma strain secretingMcAb to human p53 protein. Anti-P53 McAb which we get in the experiment can be used forstudying immunohistochemical analysis on p53 protein. Thus anti-p53 McAb made it widlyuseful in P53 detection. |
