Methylmercury (MeHg) is an important environmental pollutant whichseriously threaten the ecological environment and the people's health.As itshigh liposolubility,it can be easily absorbed into body by the food chain andtransported to the most organs,especially brain,liver,kidney.Approximately15 percent of it is located in the brain organization,resulting in the injuries ofbody systems,especially the central nervous system.This article mainlyfocuses on it's cytotoxicology and influncing mechanisms to the earlyapoptosis of the germ cells using the testing methods of health toxicologyand molecular biology.1. The cytotoxicology of MeHg to testicle germ cellsThe cytotoxicology of MeHg to the testicle germ cells was observedwith the MTT enzyme reaction colorimetry.There was significant differencesbetween the exposure group and the control group(P<0.05).It indicated thatMeHg could induce the cytotocology to the germ cells to some degree.2. The effects of MeHg on LPO in testicle germ cellsThe activities of GSH-Px decreased in the exposure group comparedwith the control group(P<0.05),and MDA content increased gradually withthe doses increasing.These indicated that MeHg could induce LPO intesticle germ cells.3. The effects of MeHg on apoptosis of germ cellsDispersed apoptotic cells in testicle germ cells were observed in allgroups treated with MeHg by hematoxylin-eosin dying,the plasm of thesecells took on light red,the nuclei shrinked and took on dark blue.Thesesuggested that MeHg could induce apoptosis of germ cells.Germ cells were strained with AO/EB to assess apoptosis.The resultsshowed that the apoptotic rate of the groups treated with MeHg was higherthan that in the control group,the apoptotic rate increased in the exposuregroup compared with that in the control group.All above further confirmedthat MeHg could induce apoptosis of germ cells,and with the doses of MeHgincreasing,apoptotic rate increased gradually.4. The effects of MeHg on germ cell cycle of male miceUsing FCM (flow cytometry) technique to examine the cell cycle, theresults indicated that the different doses of MeHg could cause the cellpercentage of different phases in cell cycle to change, with doses of MeHgincreasing, cell percentage of S, G2 phases increased gradually, and being thebiggest when in the medium dose, however, cell percentage of G0 phasedecreased with the dose increasing gradually.5. The expression of caspase-9, CytC by immunohistochemical methodCaspase-9 is one kind of enzyme that can trigger cell apoptotic cascadereaction.The results in this method showed that the membrane or plasm ofthe cells which expressed caspase-9 positively in the groups treated withMeHg took on brownish-yellow after stained.The results of image analysisshowed that the expression of caspase-9 in the experimental group wasdifferent significantly compared with that in the control group(P<0.05).Withthe increasing dose of MeHg,the expression of every group increasedgradually.It indicated that mitochondria pathway was one of the importantpathway in germ cells' apoptotic process induced by MeHg,and thecytotoxicology and LPO could increase it.The results mentioned above were very meaningful to reveal theregulating mechanism of germ cells' apoptosis induced by MeHg,and it wasalso helpful to explain the toxic mechanism of MeHg for us.
