| With the spread of the hepatitis A virus in the world,people are focus onhow to control and therapy virus .There is no specific medicine to therapy thehepatitis A except inoculate hepatitis A vaccine. There are not only commendable protection but also favourableeconomic retums for an ideal hepatitis A vaccine.There is grace anddisadvantage respectively to the hepatitis A live attenuated vaccine and thehepatitis A inactivated vaccine.The advantage of the hepatitis A live attenuatedvaccine is that only small amounts and just inoculate one time can inducepermanent and even lifetime immunization,but the safety of the liveattenuated vaccine will not be ensured if attenuation is not enough inproduce .Further more,if attenuation is excess the immunogenicity of thevaccine will be declined and potentiality to atavistic heredity will not bedepleted.Some disadvantages of the live attenuated vaccine such likeconservation time short / activity instability/ immunizing dose large has beenrestricted its exploitation in developed country. The hepatitis A inactivatedvaccine is made from attenuated strain by inactivating,although it is safe thanthe hepatitis A live attenuated vaccine ,it is complex in preparing craft andexpensive.If can find a good strain,it will be ruduced the cost and got goodeffect.The main work about this research are as follows:make the isolated hepatitis A virus adapted to the VERO cell.(1)Confirming the BL-08 strain is hepatitis A virus by total sequenceanalysis and morphological,physical,chemistrical and immumologicalmethods.(2)Confirming the virus can got ideal virus titer and antigen titer.Thebest inoculated dilution of this virus is 1:100 and the best havest time iscultured for 14 days at 35℃.To sum up ,the accomplish of this research has very considerable practicalimportance.There are so many work have to be done such as consummatepurification method ,how to impove output and animal experiment in order totestify its potencity to be ideal the hepatitis A inactivated vaccine.
