Objective The study was designed to examine the effects of fluvastatin and rosiglitazone alone or combined therapy on the expression of ABCA1 mRNA and protein in monocytes of patients with coronary heart disease in vitro. To evaluate the effect of fluvastatin and rosiglitazone combined therapy on antiarteriosclerosis.Methods Monocytes were isolated by density gradient method from blood of patientswith coronary heart disease, then divided into different group. Different group monocytes incubated with lO.Oumol/L flivastatin or 10.0μmol/L rosiglitazone for Oh, 6h,. 12h, 24h, or incubated with flivastatin or rosiglitazone at different concentrations for 24h, or incubated with lO.Oumol/L flivastatin and 10.0umol/L rosiglitazone for 24h, respectived. Total RNA of monocytes were abstracted by TRIzol reagent. The levels of ABCA1 , PPARγ and LXRα mRNA were measured by reverse transcription polymerase chain reaction (RT-PCR). ABCA1 proteins were detected by western blot. According to the difference expression of ABCA1 in each group monocyte, analyze the effects of fluvastatin and rosiglitazone alone or combined therapy on the expression of ABCA1 mRNA and protein in monocytesResults1. Rosiglitazone upregulated PPARy and LXRa mRNA expression ,it also increased ABCA1 mRNA and protein expression in dose-dependent and time-dependent manner. (Compared with control group:p<0.05)2. RT-PCR and Western Blot demonstrated that fluvastatin couldn't increase the expression of ABCA1 mRNA and protein in monocytes at different concentrations and times.
