| Skin lies in the interface between inner condition and outer environment of body, which is an important protective organ as a barrier. Hair follicle is an important and complex epidermal appendage of skin, composed of hair shaft, inner root sheath and outer root sheath. The fiberblast growth factor family comprises at least 22 structurally related growth factors. They take their biological functions by acting on FGFRs.FGFs have mitogenic effects on cells of mesodermal, ectodermal, and neuroectodermal origin, thus they are involved in physiological and pathological processes such as embryonic development, damage healing, angiogenesis, neuranagenesis, tumorigeness and so on. FGF-13 is a new member of FGF family, and murine FGF-13 may funtion importantly in the murine embryonic development, especially in the development of the nervous systems. It has been recently showed that FGF-13 protein was detectable in the bulge region throughout the hair growth cycle and may play a role in regulating the function of cells in the bulge region and basal layer of the epidermis. However, the characteristic of the role of FGF-13 in skin is unclear. Up to now, there is no report on the expression and function of FGF-13 in the follicles of skin during embryonic development and damage healing. In this study, the expression and function of FGF-13 in developmental follicles and skin during embryonic development, normal vibrissa follilce stem cells and wound follicles in vitro were obserbed,so as to illuminate the possible role of FGF-13 in the skin follicles during embryonic development and damage healing and provide the experimental base for further studying the characteristic and mechanism of the role of FGF-13 in skin. Methods:With the techniques of Immunohistochemistry (IHC), Immunocytochemistry (ICC), in situ hybridization (ISH) and Rat vibrissa follicles culture in vitro.,we studied the expression and role of FGF-13 in skin follicles during three different biological processes.First, the expression and function of FGF-13 were examined in normal vibrissa follilce stem cells in vivo and in vitro.Second, we primarily constructed a damage model of rat vibrissa follicle in vitro and detected the expression and function of FGF-13 in damage follicles during... |
