Osteosarcoma is the most common type of primary bone tumor with high malignant degree , occurred usually in adolescents . Lots of typical osteoblastic cells and tumor-like bone tissue appeared in osteosarcoma, so the malignant transformation of osteoblastic cells and the pathogenesis of osteosarcoma are well correlated. Pathogenesis and progression of tumor is a long-term process with multiple stages and multigenic alteration, having a significant complexity regulated by multiple genes and factors. Cell transformation in vitro is a good experiment method to reflect process of canceration in vivo, and is well correlated with malignant transformation in vivo. The purpose of malignant transformation experiment is to reveal what kinds of genes involved and how they work in each stage of development of a certain tumor and to provide theoretical evidence for diagnosis , prevention and treatment of it.Genes imprinting means that some genes violate the usual rule of Mendelian inheritance law and their expression is determined by the parents that contributed them. These genes are named imprinted genes, which both alleles in a heterozygote are not equally expressed. Imprinted tumor associated genes are monoallelic expressing, equivalently functional haplotype. Because such kind of genes can be inactivited by only once mutation, it increases the risk and sensitivity for tumorigenesis. The results from previous studies, which often focused on single or several imprinted genes, are significant but still insufficient for illuminating the complicated issue about tumorigenesis. A lot of uncertain imprinted genes involved in the tumorigenesis should be further defined. So investigations on expression changes of whole imprint genes in the tumorigenesis will be very important for prevention and treatment of malignant tumor .To investigate the changes of expression of imprinted genes group and correlated molecular mechanisms in the course of osteosarcoma progression, our experiment was divided into two parts.
