Osteosarcoma is the commonest primary malignant tumor in bone. The occurrence of osteosarcoma amounts to about 21% . One of the most important reasons for the metastasis of osteosarcoma is that tumor cells can survive and migrate by detaching from ECM. Loss of ECM contact could induce anoikis in nomal cells. By using the method of molecular biological technology and immunohistochemistry etc, cell surface receptor of ECM proteins can be detected. Integrins play an important role in cell growth, development and morphology. So Integrins receptors have been paid general attention in cell transformation, growth and metastasis. Because bcl-2 protein could block the anoikis signaling pathway, which has become one of the most important oncogenes in anoikis studies. With the further study of the relationship of integrins and its signaling pathway in anoikis, a new strategy will be proposed for anoikis-associated disease treatment and a kind of efficient method and a path for diagnosis of many malignant tumor.Integrins is one of the most important family members of cell adhesion molecule. It is the major receptor that can identify cells and bind extracellular matrix components, such as, collagen, fibronectin, laminins and so on. Integrins also can accept and transmit cascade signaling and regulate the biology process of cells survival, motion, proliferation etc. Integrins also play important roles in embryogenesis, tumorigenesis, invasion and metastasis, inflammation etc. The loss of integrin-mediated cell-matrix contact induces anoikis.Recently, the study of integrins and its signaling pathway is receiving more and more attention. It has been shown that protein kinase signaling pathways control anoikis both positively and negatively. Focal adhesion kinase, when activated by integrins, can suppress anoikis; phosphoinositide 3-kinase and the AKT oncoprotein may mediate the anoikis-suppressing effects of focal adhesion kinase. Conversely, the stress activated protein kinase/ Jun amino-terminal kinase pathway promotes anoikis.
