Objective:The bladder outlet obstruction(BOO) can be result in the wall of the bladder becoming thicker, and hypertrophy of the detrusor cell, then damage the function of the bladder. The critical result of BOO is the renal failure and endanger the life. For not knowing the cause of hypertrophy clearly, there is no effective method to treat and prevent the damage in clinical. The research shows the Calcineurin signal cascade plays key role in the over press induced cardiac hypertrophy and it is the important up regulate mechanism during the hypertrophy. Acoording with investigating the sub-type of the Calcineurin and measure the changes in protein level, explore the relationship between the Calcineurin and the hypertrophy .Materials and Methods: The Wistar rats are divided into experiment groups which are sacrificed 2,4,6 weeks after surgery respectly and control group. Using the partial ligation on the proximal urethral of adult rats to get the BOO model. Using morphology observation, the RT-PCR and Western blot analysis to investigate and analysis the sub-type of CnA.Results:①The increase in wet weight of the bladder after surgery is time depended and most prominent in 6 weeks.②The thinkness of the muscle layer is greater than control group. The thickness increase further after 4 weeks and decrease after 6 weeks.The fibrous tissure can be found among the muscle layer.③CnAα,CnAβmRNA can be measured at all the time point but no CnAγmRNA can be measured through RT-PCR.④CnAαexpression increases after BOO 2 weeks compare to that of control (p<0.01). Expression significantly decreases after 4 and 6 weeks. In contrast, CnB expression is unchanged obviously throughout hypertrophy development.⑤Changes in CnAαexpression agree with changes in myosin heavy chain.Conclusion:①The partial ligation on the proximal urethral of rats is a simple satisfying method to make BOO animal model.②CnAαmay play an important role...
