| Objective:Investigate the reversion effect of medroxyprogester- oneacetate on MDR1 of non-small cell lung cancer.Methods: Cancer tissue specimens and peripheral blood samples were collected from 107 patients with norsmall cell lung cancer( NSCLC),MDR1 gene sequence was amplified in total RNA extracted from cancer cell specimens and peripheral blood phocyte(PBL) by RT-PCR assay.According to the result 0f detection,some MDR1-positive patients were treated with m edroxyprogesteroneacetate and anticancer drugs( reverse group)。Other MDR1一positive patients(positive control group)and MDR1—negative patients(negative control group) were treated with anticancer drugs alone.Result:The positive rate of MDR1 in cancer tissue and PBL is 37.4%(40/107) and 34.5%(37/107) respectively (P>0.05). The objective response rate (CR+PR) was 45.9% in reverse group compared to 37.8% in positive control group,(P>0.05). The median survival time was 40 weeks in reverse group compared to 31 weeks in positive control group .(P< 0.05). The patients treated with MPA plus anticancer drugs to 160 cycle periods had increased with food intake (67.6%) , weight gain (51.4%)with 0.78±1.84kg per cycle,as well as ECOG improvement in 45.3% . According to the WHO criteria of acute toxic grade of chemotherapy, GradeⅡor more leukopenia ,was 34.0%,hemoglobin decreased 15.6% and gastrointestinal reaction18.3%.The mean recovery times of above reactions were 6.3±4.9 days,5.4士4.6 days and 1.3±1.7 days respectively.As compared with the anticancer drugs alone group,statistical diferences were significant.
