Objective In this study, cardiac myocytes of neonatal rats were stimulated with AngiotensinⅡ(AngⅡ) in the presence of atorvastatin(Ator) with varied concentrations. The protein contents of cardiac myocytes and the expression ofβ-MHC(β-myosin heavy chain) were regarded as indexes of cardiac myocytes hypertrophy. We discussed the inhibition of atorvastatin on cardiac myocytes hypertrophy and effects of atorvastatin on TLR4(Toll-like receptor 4) expression, studying statins possible mechanisms and seeking new therapy for myocardial hypertrophy. Methods Cardiac myocytes of neonatal Sprague-Dawley (SD) rats were cultured in 6 well cell culture cluster. Cells were divided into 5 groups randomly. A group: blank control group; B group:10-7mol/L AngⅡ; C group:10-7mol/L AngⅡ+ 10-5mol/L Ator; D group:10-7mol/L AngⅡ+ 10-6mol/L Ator; E group : 10-7mol/L AngⅡ+ 10-7mol/L Ator. With Collectting cardiac myocytes in 72 hours later, the method of coomassie brilliant blue was adopted to evaluate the protein contents of cardiac myocytes. The changes inβ-MHC, AT1 receptor and TLR4 expression were observed by RT-PCR.Results①The protein contents of B group were significantly higher than that of the A group(P<0.01), the protein contents of C, D group were significantly lower than that of the B group(all P<0.01), the protein contents of E group were significantly lower than that of the B group(P<0.05). The protein contents of C, D and E group gradually increased with the decrease of atorvastatin concentration.②Theβ-MHC mRNA expression of B group were significantly higher than that of the A...
