Pseudomonas exotoxin A is a single-chain cytotoxin composed of three structural domains. Domain I a (1-255 aa) is responsible for cell binding, domain II (253-364 aa) for membrane translocation enabling access to the cytosol, domain 111(400-613 aa) for the catalytic inactivation of protein synthesis, domain I b (265-399 aa) which has no clear function.Replacement PE domain I a with antibody forms immunotoxin, which is extensively used in biotherapy of tumors. Our group based on the previous research on immunotoxin, replace domain III with various apoptotic molecules to construct a new type effector to kill tumor cells. The function of domain II is to translocate the cleaved part of the toxin or other effectors from endosome to cytosol. But there are many reports to illustrate the length of PE functional translocation domain. Shorten the translocation domain and find the smallest one...
