ObjectiveImmunocontraception and immunosterility are two important problems in reproduction domain,it is key task to look for right sperm surface antigen. Sperm surface antigen has closely relation with fertilization and early development of embryo, and some antigens can ultimately leads to synthesis of special antibody .PH-20 is a traditional sperm membrane antigen,and it is a glycophosphatidylinositol(GPI)anchored hyaluronidase. The optimum pH for its hyaluronidase activity was 7.0. It is located on the sperm surface and the inner acrosomal membrane. PH-20 is a receptor for HA-induced cell signaling, and a receptor for the zona pellucida surrounding the oocyte. It has been reported that 100% effective contraception was obtained in male and female guinea pigs immunized with PH-20. Antisera from immunized females had high titres, specifically recognized PH-20 in sperm extracts, and blocked sperm adhesion to the egg zona pellucida in vitro. Immunized females, mated at intervals of six to fifteen months after immunization, progressively regained fertility.So we can see that long-lasting and reversible contraception was obtained in male and female guinea pigs immunized with PH-20. PH-20 is an important candidate for contraceptive vaccine.DNA vaccine is one sort of new vaccine and developed from 1990.
