Background and purpose:Many kinds of endogenesis neuropeptides have been isolated and studied since P substance was purified in 1931. They have various functions on regulation to nerve system. Neuropeptide, a new drug resouce, is widely attaching importance.Our group isolated a new bovine neuropeptie, acidic peptide (AP), from bovine brains. The acidic peptide is composed of glutamic acid. It had been shown that the new acidic peptide has obvious biologic activity.It had proved that acidic peptide had a good therapeutic effect in the mice with viscular dementia and AD model rats. Acidic peptide increased abilities of learning and memory of them, and induced an increase of activity of superoxide dismutase (SOD), and decreased the production of malondialdehyde (MDA). Acidic peptide also obviously reduced the formation of nitric oxide and nitric oxide synthase. This prevented from the damage of rat' s nervous system, and promoted the rehabilitation of damaged brain. It had been found by experiments that acidic peptide had anti-mutation. Acute toxicity test indicated that the safety of acidic peptide was very good. Its oral tolerance dosage exceeded 5g/kg.In order to observe long-term toxicity, possible target organ, reversibility after lesion and dosage for clinical use, studies of long-term toxicity and carcinogenicity had been done.
