Objectives: High lipid diet induced AS rabbit model is used to further observe the antiatherosclerotic effect of rhubarb ethanol-extract (REE) and its possible mechanism is explored, in order to provide more accurate theoretical and experimental foundation for using more reasonably and the development of new antiatherosclerotic drugs with rhubarb.Method: Thirty healthy male white rabbits were divided randomly into five groups. The rabbits in control group were fed with common forage. The rabbits in model group were fed with high lipid forage. The rabbits in three different dose REE groups were fed with high lipid forage and treated with different level REE. In the before experiment, in the middle period and at the end of experiment, the blood samples were collected. Some blood components, such as total cholesterol (TC), triglyceride (TG), high density lipoprotein cholesterol (HDL-C), low density lipoprotein cholesterol (LDL-C), were tested. Periodically measured the rabbit weight during the period of feeding experiment, and compared increase heavy variety for average days with rabbit. The rabbits were killed at the end of tenth week, the AS plaque areas of aorta were measured and the percentage of AS plaques covering aortic intima (PA) was calculated. The pathological changes of coronary arteries were observed with microscope and the incidence of atherosclerosis of small coronary arteries was calculated. Liver fatty degeneration degree and liver coefficient were measured and compared, and researched REE protected liver fatty degeneration function.Results:1. TC Serum total cholesterol (TC) concentration in model group was significantly higher than that in control group (P<0.01), which indicated the model was successfully made. TC in the three rhubarb groups were not significantly lower than that in model group beside the REE III rhubarb group in the middle period (P<0.05).
