Background Oxidized low density lipoprotein(Ox-LDL )is one of the major players in the genesis and development of atherogenesis. Recently, several reports have demonstrated that the level of circulating Ox-LDL is high in patients with atherosclerotic disease. Because of a lack of a sensitive method to specifically detect circulating Ox-LDL level in China, the clinical importance of circulating Ox-LDL is poorly understood. The use of double monoclonal antibody sandwich method is one of the integral parts of a specific and sensitive diagnostic strategy.Objective To prepare monoclonal antibodies (McAbs) against Ox-LDL, development of double monoclonal antibody sandwich method for detection of human serum Ox-LDL level and its primary clinical application.Methods LDL has been obtained after serum ultracentrifugation ( 1.019 ~ 1.063g/ml) , and then been oxidized with 20μM CuSO4 for 8 h ( Ox-LDL) .
