Objective:The present study was to investigate the effect of valproate (VPA) on testosterone secretion and steroidogenesis acute regulatory protein (StAR) , cholesterol side-chain cleavage enzyme (P450scc) and cytochrome P450 aromatase (P450arom) mRNA expression in porcine thecal cells in vitro and the effect on steroidogenesis and gene expression of CYP17, P450SCC and StAR in human granulosa cells from both normal women and patients with polycystic ovaries. The aim of this study was to demonstrate the mechanism of the VPA resulting in PCOS like syndrome, and the possible pathyway of the environmental chemicals relating to enovulation.Methods:Porcine thecal cells and human granulosa cells were isolated and cultured with DMEM-F12 (1:1) medium and treated with various concentration of sodium valprovate (0, 100, 250mg/L). The culture media were collected after 48h for testosterone measurements by ELISA, CYP17, P450SCC and StAR mRNA in thecal cells were detected by Real-time RT-PCR. Progesterone and estradiol were detected by standard radioimmunoassay, and StAR, P450scc and P450arom mRNA were detected by real-time RT-PCR in human granulosa cells.
