Objective To study differentiation situation of blood B lymphocyte cell and its subsets (Naive B cells, memory B cells and plasmablasts) in peripheral blood of patients with rheumatoid arthritis (RA) by two color flow cytometry.Furthemore, and to analyze theirs relationship with autobodies and clinical manifestation.Methods Blood samples were obtained from 60 RA patients. All patients fulfilled the criteria for RA classification proposed by the American College of Rheumatology (ACR). They were divided into three groups which were active (21), inactive (19) and refractory (20) RA, by clinical manifestation and drug reaction. Meanwhile 32 SLE patients who fulfilled the ACR criteria for SLE classification and 24 healthy individuals were used as controls. B Cell surface markers(CD19,CD27) expression with immunofluorescence staining was determined by flow cytometry. Frequence of Naive B cells (CD19+CD27- ), memory B cells (CD19+CD27dim) and plasmablasts (CD19+CD27high) was analyzed. In addition, autobodies such as: RF, anti-type â…¡ collagen (Câ…¡) antibody and anti-cyclic citrullianted peptide (CCP) antibody were collected. Clinical manifestation such as morning stiffness, tender joint count, swollen joint index, HAQ et al were recorded. Laboratory items such as: erythrocyte sedimentation rate (ESR) , C-reactive protein (CRP) et al were tested. B lymphocyte subsets in periphral blood of patients with RA and theirs relationship with clinical manifestation and autoantibodies were investigatied.
