Abstract/Summary:
Objective:To investigate the effect of adenovirus-mediated gene theapy with vascular endothelial growth factor (VEGF) to treat compromised skin flaps in the rat. In addition,this study determine the effects of different injection layer .Methods:Fourty Sprague-Dawley rats weighing 350-400g wereused in this study .the rats was randomized into four groups of ten rats each .A dorsal flap (8cm X 2cm) in full thickness with the pedicle remaining attached at the posterior end was elevated in the 40 rats and received the following treatment: group Ad-VEGF1 (n=10):subdermal Ad-VEGF injection into the flap (10 Pfu/ml ) after flap elavation ;Group Ad-VEGF2(n=10) topical injection onto the recipient bed (10 Pfu/ml) after flap elavation ; GroupAd-VEGF3(n=10):subfascial injection into the recipient bed (10 Pfu/ml) after flap elavation ;Group Control (n=10) :without any treatment after flap alavation .Results :after7 days recovery , the mean percentage of surviving flap area was (74.56±1.70)% in group Ad-VEGF1,(74.12±1.17)% in group Ad-VEGF2 ,( 72.52±1.18)%in group and Ad-VEGF3 (52.24±1.87)% in group control .there was a significant increase in the percentage of the survival area in the flaps of the group Ad-VEGF1,Ad-VEGF2 and Ad-VEGF3.compared with the group control .No significant difference were found among the gene therapy group .but the mean percentage of surviving flap area of grouped-VEGF1 and Ad-VEGF3 high than group Ad-VEGF2.Immunohistochemical staining showed that VEGF was expressed in the survival flap injected with Ad-VEGF .Histological analysis showed that more granulation tissues and angiogenesis were observed in the gene therapy group .
