In recent years, the development of traditional Chinese drugs(TCD) has been some impeded because of the adverse drug reaction (ADR) . Therefore, it is an urgent task to evaluate the objectivity of its ADR, establish the rational use strategy and ensure the security and validity of TCD. Radix et Rhizoma Rhei has been used commonly and its ADR was reported occasionally. It is possible to affect other anthraquinone-containing drugs like Radix Polygoni Multiflori, Alose and so on while the ADR couldn't be well solved.The processing and compatibility are the main characteristics and forms in the application of TCD. The different processed products and Dahuangzhechong pills were chosen and the chemical and bioactive variation of processing and compatibility of Radix et Rhizoma Rhei were investigated in this paper.Chemical technique was used to determine the content disparities of anthraquinones in different Radix et. Rhizoma Rhei samples. A series of fingerprints of the samples were established and analyzed. Microcalorimetry was used to investigate the interaction between different samples and different experiment models. The energy transfermation and parameters changes were observed in the course. Pharmacogical and toxicological tests were used to evaluate the difference of action and toxicity of different samples. The scientific mechanism of the processing and compatibility of Radix et Rhizoma Rhei.hzd been probed.The results suggested that it would be safe if Radix et Rhizoma Rhei was used reasonably. The processing and compatibility had caused the chemical constitutes changed especially the conjugated anthraquinones. It also showed that there was an obvious correlation between biothermodynamic parameters and pharmacological data. Biothermodynamics is a feasible means to evaluate the bioactivity of TCD.The chemistry-bioactivity model based on processing and compatibility can be broadened out to TCD which contains toxic components like anthraquinones and others.
