Purpose: Leiomyosarcoma(LMS) of human soft tissue accounts for 5 ~ 10 % in soft tissue sarcomas, which has complicated histology and bad prognosis. Few predisposing or etiologic factors have been recognized for this disease. So there is little progress in its diagnosis and treatment now. We establish tissue microarray in 20 LMS, 12 leiomyomas ( LM) and 9 normal smooth muscle tissues. Then we detect several associated proteinum and a gene of LMS with immunohistochemistry(IHC) and hybridization in situ (ISH) techniques. Including osteopontin (OPN), Ki-67, P53, P16, demin and α-SMA. Researchers find that OPN plays an important role in pathogenesy, invasion and metastasis of tumors now. Nevertheless, we now know little about how OPN expressing in LMS and its significance. So we detect OPN expression, using IHC and ISH, and analyze its significance. We also study Ki-67 expression in smooth muscle tumors and analyze the difference between benign and malignant ones and its significance. Different researchers reported different results about how P53 and P16 expressing in LMS, we gain our results without variability which is induced by the intra- and inter-run with the use of tissue microarray...
