Objective To investigate the role of mRNA expression of imprinted tumor suppressor gene ARM (A ras homologue member Ⅰ) in tumorigenesis and progression of ovarian cancer. To explore the expression of ARHI in ovarian cancer cells and the effects of HDACIs (histone deacetylase inhibitors)on the growth of cells and the expressiong of ARHI.Methods1,All the ovarian tumor were placed under -80℃ after removing from body. mRNA expression of ARHi gene was evaluated in normal ovary(n=10),ovarian begin tumor(n=13),ovarian borderline tumor(n=5),ovarian cancer(n=33) by RT-PCR. The mRNA expression of ARHI was compared with clinicopathological features of ovarian cancer.2,After the secondary culture of ovarian cancer cell lines(SKOV3,A2780, COC1, OC3),inhibition of cell growth induced by TSA(Trichostain A) or CDDP(Cisplatin) at different concertration was detected by methyl thiazolyl tetrazolium(MTT). And the effects of HDACIs on the expression of the ARHI in four ovrian cancer cell lines was detected by RT-PCR.Results1,Relationship between the mRNA expression of ARHI gene and pathobiological...
