The Gene Of Hepatocyte Growth Factor To Transfect The Chondrocytes Of Rabbit Condylar And The Research Of TMD Cured By Chondrocytes Transfected

Objective To study the main biological chacteristics of chondrocytes with gene-transfected by HGF (hepatocyte growth factor) cDNA Labeled with GFP (green fluorescent protein) in vitro . and to investiage the effection of TMD cured by chondrocytes transfected. Methods The cells were derived from rabbit condylar ,and were cultured to the second generation in vitro,then the cells were infected by HGFcDNA loaded with liposome,then the shape of cells were investiaged under inversion microscope ,the capacity of cell proliferation was tested by MTT,The differences of the type 11 collagen were tested by immunohistochem between the transfection group and comparison group ,and so on the mRNA levels were tested by in situ hybridization of the type II collagen between the transfection group and comparison group . The chondrocytes transfected was counted ten times under fluorescen inversion microscope, made the average,It was the percent of the chondrocytes transfected. We made the animal model of TMD by rejecting collage enzyme to joint cavity of TM,and check the result of curing by tissue slice of joint. Results The chondrocytes of the prime generation were anchorage-dependent cells,the cells anchored when it was the third day, the bottom of bottle was full of chondrocytes when it was the tenth day ,The shape of cells was polygonal or triangular; the GFP was investiaged in the chondrocytes transfected under fluorescence inversion microscope after 24h,the condylar chondrocytes transfected by HGF could keep the main biological chacteristics of chondrocytes more time than comparison group,the cells of the first generation which were transfected became longer than comparison group,it got back its shape after about two generations. HGFcould accelerate the speed of cell proliferation, improved cell proliferation. The expression of collagen II is stronger than the comparison group on the seventh generation by immunohistochem.
