objective: To research the influence of retinoic acid on the differen tiation of cord blood stem cells and to approch the condition of intro ducing cord blood stem cells to differentiate to neuron-like cells in vitro , which provide experimental fundament for the study whether transplanted cord blood stem cells can repair injured spineMothods: hMSCs were separated from cord blood by density gradient centrifuging and cuthered in DMEM with 20%FBS and amplificated at the same condition, then take the 5th generation of amplificated hMSCs to have an innoculation in the culture plate and divid into 3 groups on the basis of concentration of RA and the induced time then add inducer RA to the culture medium, and cultivate at 37℃,5%CO2 in satured humidity then observe the change in morphology and histology. After 24h, 48h and 72h, to detect the neuron symbol by immunohistochemical method and observe the sample under microscope, count 10 unwraped eyeshot randomly and calculate the positive cell percentage of the neuron symbol neuron-specific enolase (NSE), neurofilamentproteinM (NF-M), gliad fibrillary acidic protein (GFAP) of every groups, denoting as ( x|- ± s)%. to compare the role of retinoic acid at different induced time in the induced process .
