Objective: To investigate the feasibility and efficacy of transplantation of mesenchymal stem cell and the electrophysiological mechanism for treatment of myocardial infarction in rat.Method1.To isolate, purify, amplify and culture the mesenchymal stem cell: The bone marrow of Wistar rats were collected in aseptic condition.The bone marrow stem cell were cultured and amplificed in culture flask.2.To induce,identify and mark the bone marrow stem cell: The bone marrow stem cell were induced by 5-azacytidine for 24 hours.The actin and troponin were identified by immunohistochemistry.The plasmid p-GFP as a marker was transfected into the induced cells.3.To establish the model of myocardial infarction and transplant the cells to the infarcted myocardium: 30 Wistar rats were anesthesized by Chloral Hydrate. The free wall of left cardiac ventricle were frozened by liquid nitrogen. All the rats were divided into two group randomly.Every group had 15.The cells were transplanted into the infarcted sites in infarcted group while the culture fluid was transplanted into the corresponding sites in the control group.
