Purpose : To cultivate rabbit bone marrow mesenchymal stem cells (MSCs) in vitro and induce it to differentiate into cells with Schwann cell characteristics. Stably introduce enhanced green fluorescent protein gene( EGFP ) to it by retroviral vector. We investigate their biological characteristics and ability to be induced to differentiate into Schwann cell-like . The intensity and time of luorescent light expression is also observed . So as to approach the feasibility of MSCs be applied to induce nerve regeneration in bioengineered nerve conduits and whether green fluorescent protein is a stable morphological marker for MSCs transplanted inbioengineered nerve conduits.Methods: MSCs were isolated from rabbit bone marrow by density gradient fractionation and adherence to plastic flasks. Then MSCs was treatedwith beta-mercaptoethanol followed by retinoic acid and cultured in the presence of forskolin , basic-FGF , PDGF and heregulin , changed morphologically into cells resembling Schwann cells and expressing S-100.Meanwhile packaging cells PT67 were transfected by pLEGFP-N1 retroviral vector , and MSCs were transduced by viral supernatant...
