Objective: There are many methods to repair peripheral nerve defects. The most useful one is free nerve autograft especially with blood supply . But the resources are 1.0imited and the method can not adapt to long piece or mutipl-nerve deficiency. Xenoma nerve is another useful nerve conduct that are sufficiency and preserve. But allograft leads to immunological rejection. The conduct made of non-nerve material or biomaterial and artificial nerve are still in animal research. We learn that the end-to-side anastomosis can change injury from high position to low position. Patients taken this operation cost less and suffer less pain. We study the effect of end-to-side anastomosis compare to free nerve with blood supply autograftMethods: Twenty adult Sprague-Dawley rats (200- 250 g) were establish free nerve with blood supply autograft model and the other hindlimb taken end-to-side anastomosis. Exposed sciatic nerve , we abscise 1cm long from 0. 5cm level after diverging fibula nerve. Pay attention not to lib mesentry of nerve and not to injury vasa nervorum .Then suture adventitia of the free nerve by 11-0 thread . We abscise the other sciatic nerve at the same level. Suture the distal end of fibula nerve mesentry to tibia nerve mesentry by 11-0 thread in 45' Culture them separately and observe hindlimb paralysis and sensation recovery. After 6 month, we abscise distal of fibula nerve. Then measure the dimension and quantity of axon by methylamine , chromotrope 2R brilliant green and HE staining . We use transmission electron microscope to measure the thickness of myelin sheath and the area of medullated nerve fibers.
