Purpose To detect P63,K3 expression in human limbus, to approach the possibility of P63 acted as the marker of human limbal stem cells.Method 8 fresh normal human corneal tissues were obtained from 8 donors who suffered serious rupture of globe.we obtained corneal tissues through doing evisceration of eye.24 corneal tissues were obtained from 24 donors who were diagnosed limbal stem cell deficiency(LSCD), The diagnosis of LSCD is based mainly on clinical signs. we obtained these corneal tissues through doing evisceration of eye.after surgery The tissue specimens were immediately fixed in 10% phosphate buffered formalin for 1 day, and embedded with paraffin. Paraffin sections (4μm thick) were then cut for in Immunohistochemical Staining experiments.Results p63 protein was immunodetected only in the nuclei of limbal epithelial basal layer, but not in most limbal suprabasal ,paracorneal and corneal epithelial cells.in corneals of local limbal stem cells deficiency,because there are probably some health limbal stem cells in limbus, p63 protein was immunodetected only in the nuclei of limbal epithelial basal layer,but the expression is reduced.in corneals of diffuse limbal stem cells deficiency,with the extensive loss of limbal stem cells, p63 protein was not immunodetected in whole corneal.K3 protein was not immunodetected only in the nuclei of limbal epithelial basal layer, but in limbal suprabasal ,paracorneal and corneal epithelial cells. in corneals of local limbal stem cells deficiency, K3 protein was...
