Objective: Ligation of CD40 paly an important role in the B-cell tumor immunotherapy. We studied the relationship of Ligation of CD40 with other apoptosis associated factors and investigated mechanisms of the biological effects mediated Ligation of CD40 by soluble recombinant human CD40 ligand (srhCD40L) on human Burkitt's lymphoma CA46 cells. Metheds: By using Trypan blue exclusion and flow cytometry, cell apoptosis and the biological effects mediated Ligation of CD40 by srhCD40L on human Burkitt's lymphoma CA46 cells were studied. By using western blotting, The protein expressions of Survivin,c-Myc,hTERT,NF-ΚB,Bcl-2,Casepase 3 on CA46 cells dealed with Ligation of CD40 by srhCD40L were investigated.Results: Ligation of CD40 by srhCD40L resulted in cells growth inhibition and apoptosis of CA46. Ligation of CD40 by srhCD40L could inhibit the protein expressions of survivin, c-Myc, hTERT, NF-ΚB, Bcl-2 and up-regulat the protein expression of Casepase-3 in a time-dependent manner.Conclusion: Ligation of CD40 by srhCD40L inhibits human Burkitt's lymphoma cell line CA46 cells proliferation and induces apoptosis. Down-regulate the protein expression of survivin, c-Myc, NF-ΚB, Bcl-2, up-regulate the protein expression of Casepase-3, and inhibition of telomerase, may play important roles in the apoptosis of CA46 cells induced by Ligation of CD40 by srhCD40L.
