Objective: To investigate the effects of irbesartan on transcription activity of peroxisome proliferators-activated receptor v in the kidney of diabetic rats which treated with irbesartan or irbesartan +pioglitazone , and further investigate the possible mechanisms.Methods: Male Sprague-dawley rats were randomly divided into five groups as followed(10 rats in each group):normal control group (NC) , diabetic control group (DMC), irbesartan(50mg/Kg. d)-treated diabetic group(DMI), pioglitazone(5mg/Kg. d)-treated diabetic group(DMP), irbesartan(50mg/Kg. d ) + pioglitazone(5mg/Kg. d) diabetic group(DMIP). Diabetic rats were induced by intraperitoneal injection of streptozotocin in the dose of 65mg/kg. Treatment lasted 12 weeks. Body weight and blood pressure were measured, and serum glucose , serum cholesterol, serum triglyceride , serum creatinine, blood urea nitrogen , 24-hour urine protein excretion were determined in week 12. And then all the animals were killed. Glomerular extracellular matrix was measured by PAS staining; expression of p-ERK in glomeruli was detected by immunohistochemistry; mRNA expressions of PPAR-γ and A-FABP in renal cortex were determined by RT-PCR assay, mRNA expression level of A-FABP was regarded as the index of PPAR-Y transcription activity.Results: (1) Renal lesions presented in diabetic rats after 12 weeks since STZ injection.
