Objective: Ischemia reperfusion injury of the lung frequently occurs in many clinical conditions, especially during lung transplantation. Reperfusion injury after lung transplantation continues to contribute to increased morbidity and mortality, so it is necessary to better understand the mechanisms of lung ischemia reperfusion injury and investigate one more reliable strategy to reduce the injury. This experiment was carried out using the model of rat lung ischemia reperfusion injury in vivo. The soluble intercellular adhesive molecule l(sICAM-l) concentration in plasma, the interleukin 10(IL-10) and the contents of myeloperoxidase(MPO) in lung tissue were measured. The pathological changes of the lung tissue were observed to explore the effects of ulinastatin(urinary trypsin inhibitor, UTI) on ischemia reperfusion injury of rat lung in vivo.Materials and Methods: Forty-eight male Sprague-Dawley rats were anesthetized with an intraperitoneal injection of sodium pentobarbital (30mg/kg). The animals were shaved and endotracheal intubation with a 14 gauge tube, and ventilated with a rodent respirator ventilator. With supplemental oxygen at a rate of 60 cycles per minute, a 1:1.5 of I/E ratio and a maximum of 0.03 to 0.04 M Pa peak pressure were used. All animals received 0.04 mg/kg of intramuscular atropine after being anesthetized. The chest was opened via a left thoracotomy through the fourth or fifth intercostal space, after the left pulmonary hilum including all neural, vascular, lymphatic, and connective tissue was stripped, a ligature was placed through, and the ends of the tie were threaded through a small plastic tube to form a snare for reversible left pulmonary hilum occlusion. Heparinization was maintained during the experimental period with a bolus injection of 100U/kg sodium heparin via the dorsal penile vein.
