Objective: To observe the expression and changes of nuclear factor-KappaB(NF-κB) in mice acute lung injury(ALI) induced by lipopolysaccharide (LPS); investigate its mechanism in ALI and the therapeutic effect of BN52021 on ALI.Methods: Fifty healthy Kuenming mice were randomly divided into two groups: L group and B group(n=25 in each group). LPS (15mg/Kg) was intraperitoneally injected in L group ;BN52021 (20mg/kg) was intraperitoneally injected in B group 5 min before LPS injection. At 0,1,3,9 and 12h after LPS injection, arterial partial pressure of oxygen(PaO2) and lung wet/dry ratio(W/D) were recorded; Expression of NF-κB p65 was detected by using Western Blotting; the levels of tumor necrosis factor-α(TNF-α) and interleukin-10(IL-10)was determined by using ELISA;the expression and distribution of NF-κB p65 in mice lungs was detected by immunohistochemistry, at the same time pathological changes were examined under optical microscope.
