Objective: To investigate the cultivation and biological character of corneal epithelial cells(EC) and oral epithelial cells planted on amniotic membrane (AM) as a substrate trying to find out satisfactory methods and rich material for ocular surface reconstruction.Methods:⑴Denuded AM was obtained by using equivalent mixture of 0.25%trypsin and 0.02%EDTA and was adopted as a substrate with intact AM.⑵Rabbit limbus tissue pieces and corneal epithelial cells obtained through digestion were planted on denuded AM and intact AM respectively with DMEM/Ham,s-F12 as culture medium.⑶The cultivated corneal epithelial sheet was examined by inverted microscope,HE staining pathological section and electron microscopy (EM) compared with normal rabbit cornea epithelium.⑷The cultivated epithelium sheet was also immunohistochemically labeled with AE5 monoconal antibody reacted with SP Histostain-plus kits and DAB developer to identify the expression of cytokeratin 3(CK3).⑸Oral epithelial cells(EC) were planted on...
