Objective: To enhance survival rate and purity of primary cultured hippocampal neurons by improving traditional hippocampal neurons culture method, study electrophysiological properties of the cultured neurons.Methods: Neonatal rat hippocampal neurons were isolated and cultured in the serum-free B27 supplemented culture media. The process of hippocampal neurons growth and differentiation was observed. Neurofilament protein (NF) was detected by immunofluorescence staining to indentify neurons and to calculate neuron ratio.
