Purpose: To explore the mechanism of molecular chaperone function ofα-crystallin; To further illuminate etiopathogenisis of cataract; To supply experimental data and theoretical knowledge for drug treatment of cataract.Methods: Citrate synthase (CS) was denatured by guanidine hydrochloride and chosen as the target protein.α-crystallin of different concentration (0nM, 50nM, 150nM and 300nM) was added into denatured CS. Spectrophotometer was used to record the relative light scattering rate of each concentration to assess the aggregation of CS in 6 minutes.α-crystallin of different concentration (0nM, 150nM, 300nM and 600nM) was added into denatured CS induced by guanidine hydrochloride and spectrophotometer was also used to record the absorbance of each concentration to assess the activity change of CS in 2 hours. On the other hand, CS, guanidine hydrochloride andα-crystallin of different concentration (0μM, 1μM and 2μM) were incubated together, and the remaining activity of CS in each sample was detected in 30 minutes by spectrophotometer.
