Objective: To investigate the effects of simvastatin on the cell proliferation and apoptosis of K562 cells and to elucidate its molecular mechanisms.Methods: 1. K562 cells had been treated with various concentrations of simvastatin for different time, and then MTT assay was employed to evaluate the effects of simvastatin on the proliferative inhibition of K562 cells. 2. Annexinâ…¤-FITC/PI staining, light microscope and agarose gel electrophoresis were performed to confirm the apoptosis of K562 cells incubated with simvastatin for different time, and the apoptotic effect of the combination of simvastatin and MIT after a 72h incubation was analyzed by Annexinâ…¤-FITC/PI staining, and cell cycle of K562 cells treated with simvastatin for 24h was assayed by flow cytometry. 3. P53 and PCNA protein expression levels in K562 cells treated with simvastatin for 48h were measured by immunohistochemistry method. 4.
