Background Mitogen-activated Protein kinase(MAPK) play an important role in cell signal transduction. It is a common pathway and convergent Point in cell signal transduction, in which relevant signals out of cell are transmitted to cell nuclear to affect the transcription and expression of genes. Recently, it has been thought that MAPK signal Pathway is involved in the regulation of cell proliferation and apoptosis. An imbalance between cell proliferation and apoptosis may play a crucial role in the genesis and development of skeletal muscle Movement injury. At present, the role of each number of MAPK family in the genesis and development of skeletal muscle injury has been attached great importance to.Objective TO study the role of p38 in the genesis and development of skeletal muscle Movement injury , we investigated the expression of activation p38 , apoptosis index and Z-line damaged in rat's gastrocnemius muscle after different loading treadmill, respectively.Method There were 78 SD rats specimens, CON group in 6;normal exercise group in 24,moderate exercise group in 24,exhaustive exercise group in 24, respectively. They were divided into 4 grade by movement time 1d, 3d, 7d, 14d.Western-blot was used to detect the activation of p38, PI (Propidium iodide) was used to detect cell apoptosis; electron microscope was used to detect Z-line damaged. One-Way ANOVA and Spearman rank relation analysis in SPSS 10.0 and Origin software were used as the statistic methods, p values less than 0.05 were considered statistically significant in bilateral.Results Z-line damaged were highest in D3 and significantly higher in A, B3, C3 compared to D3, respectively(P<0.05).The gastrocnemius cell apoptosis index was not obvious in normal exercise groups, respectively (P>0.05); The gastrocnemius cell apoptosis index was statistically significant in moderate exercise groups and exhaustive exercise groups, respectively (P<0.05).The gastrocnemius cell apoptosis index was highest in exhaustive exercise group after exercising.
