In order to study melanin content and distribution in vitiligo skin, the reflectance and fluorescence for vitiligo were studied in this paper.Firstly, a short introduction to the study history of vitiligo, melanin and spectroscopy used in the diagnosis of skin diseases were briefly outlined. The skin structure, skin physiology and diseases were also given.Then, the principle and application situation of spectral technique in medical field were in detail. Meanwhile, the method computerizing reflectance and fluorescence to study melanin content and distribution in skin was advanced.Thirdly, the reflectance and fluorescence spectra of trichrome vitiligo skin were taken using reflectance-fluorescence system. On the basis of the skin optical model advanced by Prof. Zeng, a proper model for the object we used was developed.Afterwards, compared the spectra taken by reflectance-fluorescence system with that calculated by Monte Carlo simulation, rectifying melanin content and distribution in the model, to make the spectra calculated by Monte Carlo simulation agree with the spectra taken by reflectance-fluorescence system in general.Finally, the impact of blood content on skin spectrum was studied by using Monte Carlo simulation.Vitiligo skin, especially trichrome vitiligo skin is an ideal in vivo model for studying the effect of skin melanin content and distribution inn skin spectroscopy properties. Research methods developed here, including the spectral ratio method, the method of developing and modifying melanin content in the optical model, and Mont Carlo simulation may also be useful for other non-invasive optical studies of skin and the skin diseases with pigmentation or varices.
