Objective: To identify the location of the lung cancer associated protein N35 in subcellular structures by laser scanning confocal microscope. The distribution of protein N35 in different phages of the cell cycle was also observed to learn more information about clinical potential application.Methods : The cells culture, ELISA, immunoflourescence , laser canning confocal microscope were used by the monoclonal antibody N-35 as the immunoprobe to detect the subcellular localization of tumor associated protein. Results : Tumor associated protein N35 obviously distributed on the nuclei significantly more than other subcellular structures. It distributed in the live lung cancer cell GLC-82, but not in the protein ingredient of the normal human lymphocyte, the normal human lung cell and human skin fibroblasts, Tumor associated protein N3S was located on the centriole of the chromosomal domain at the S-G2 phage.Conclusions : The tumor associated protein N35 might be a tumor cell growth regulator, expressed only in the cancer cells and related with the proliferation of cancer cells. It might be one of the tumor markers. It could by used clinically in earlier diagnosis of lung cancer, and evaluation therapy effect, investigationrecurrence, predict metastasis and theory prognosis.
