It is the one of the milestones in 20th century that the immunosuppressor is widely used in clinic. The wide application of the immunosuppressor can decrease the rate of the acute transplantation rejection. However, the fact that the immunosuppressor can lead to toxic or side effect as well as the allograft injury inhibits the increase of the long-term survival rate of the patient as well as the allograft. It was reported that the activated T cell is one crucial cell which can lead to transplantation rejection. The rate of the transplantation rejection as well as the toxic or side effect of the drug can be decreased if the activated T cells are selectively killed while the quiescent T cells are still remained.The activated T cell can induce the expression of the cytotoxic T lymphocyte antigen-4 (CTLA-4) molecule. The engineered immunotoxin upon the recognition of CTLA-4 molecule can kill the activated T cell specifically but remain the quiescent T cell.
