Objective Mucus hypersecretion is a distinguishing pathophysiological feature of many respiratory diseases. MUC5AC is the most important mucin which has been researched.We founded rat models with airway mucus hypersecretion induced by aerosol acrolein inhalation to investigate the effect of intragastric administrations of sildenafil on airway mucus secretion and discuss the potential mechanisms of sildenafil on mucus secretion. Methods Rat models with airway mucus hypersecretion were established by aerosol inhalation with 3.0 PPM Acrolein for 6 hours per day. The experimental group was administered intragastric administration of sildenafil (25mg/Kg ) while the control group was administered intragastric administration of saline. BALF and lung specimens were acquired for detectionat at the 14th and 28th day . TNF- α in BALF was detected by ELISA. The expression of mucin was detected by AB-PAS staining. Intracellular Mucin5AC was detected by immunohistochemical staining. The average optical density of positive staining of Mucin5AC was analyzed by image analysis technique.Results Intragastric administration of sildenafil significantly inhibited the generation of neutrophil cell induced by aerosol in BALF (P<0.05 respectively). however, there was no significant decrease of lymphocyte in BALF (P>0.05 respectively). Sildenafil also significantly inhibited the generation of TNF- α in BALF which induced by aerosol (P<0.05 respectively).At the 14th and 28th day wih stimulation with acrolein ,the AB-PAS positive staining and immunohistochemical positive staining were obviously up-regulated in bronchi with a time-dependent manner ,comparing with...
