ObjectiveThe purpose of this study was to detect the expression of β2 adrenoceptors on human bone marrow mesenchymal stem cells (hMSCs) and investigate the effect of β2 adrenoceptor agonist/antagonist on the mRNA expressions of BMP2 and cbfa1 during the process of osteogenic induction (OSI) of hMSCs in vitro. MethodThe hMSCs of a healthy adult volunteer bone marrow were isolated by the method of density-gradient centrifugation. The hMSCs were affirmed by flow cytometry(FCM) and OSI. At the third passage, the β2 adrenoceptors mRNA on hMSCs was detected by Real time Quantitative PCR. Then hMSCs were divided into three groups by randomization: A (β2 adrenoceptor agonist group), B (β2 adrenoceptor antagonist group) and C (normal control group). During the process of hMSCs to differentiate toward osteoblasts in vitro, A and B groups were given 1 μmol/L, 5μmol/L and 10μmol/L Salbutamol (β2 adrenoceptor agonist) and ICI-118551 (β2-adrenoceptor antagonist) respectively. The mRNA expressions of BMP2 and cbfa1 were measured by PCR at 48 hours, 72 hours and 96 hours respectively.
