Objective: To study the relationship between the relative quantification of HER-2/neu DNA and biological behavior of breast cancer. Exploring the feasibility of judging HER-2/neu protein expression in tissue by serum HER-2/neu DNA. Methods: Total of 50 untreated serum specimens obtained from patients with breast cancer were studied of HER-2/neu DNA by fluorsesc-ence quantitative polymerase chain reaction (FQ-PCR).Of them, 30 were HER-2/neu protein positive in tissue by immunohistoch-emical,20 were HER-2/neu protein negative. Pathological type: 39 invasive ductal carcinoma, 11 invasive lobular carcinoma. As control, 20 serum samples from patients with benign breast tumor were also analysed. Results: 9 of 50 cases with breast cancer for serum HER-2/neu DNA were amplified. The positive rate wasl8%(9/50). 9 cases were all with HER-2/neu protein expression in tissue. The positive rate in them was 30(9/30); Serum HER-2/ neu DNA was significant difference among HER-2/neu protein positive in tissue group, negative group and control group (P<0.05) .There was no significant difference between HER-2 /neu protein negative group and control group (P>0.05) , and no significant difference with age, tumor size, pathological type, ER, PR (P>0.05) .Significant difference of serum HER-2/neu DNA was also found among lymph node metastasis ≥3 group, <3 group...
