OBJECTIVE To investigate the pathology of the cerebral aneurysm and the expression and activity of i-NOS,MMP-2 and MMP-9 in the cerebral aneurysm wall and normal artery. Try to explain the difference of i-NOS, MMP-2 and MMP-9 level between two groups, and find their meanings to the pathophysiological mechanisms of cerebral aneurysms.METHODS The cerebral aneurysm walls were excised from 20 patients undergoing craniotomies for aneurysm clipping and 15 normal cortical arteries were prepared. Some of then were examined by light microscopy (Hematoxylin and i-NOS, MMP-2 and MMP-9 immunohistochemistry stained) to investigate the pathology. And the i-NOS,MMP-2 and MMP-9 activity in the other specimens were detected using spectrophotometry and substrate gel zymography.RESULTS Under light microscopy observation, the specimens from the cerebral aneurysm group lost their normal structure. Intimal fibrous thickening and disappearance of the internal elastic lamina can be seen in all of the cases. Lymphocytes and macrophage infiltrated the entire aneurysmal walls. Atherosclerotic lesions in different stages were common. The strong express of MMP-2,MMP-9 and iNOS were seem in all the cerebral aneurysms, and they were express major in the inflammatory reaction area .In the control group, the expression of MMP-2,MMP-9 and iNOS is none or weak.
