PURPOSE: To study the effects of Exendin-4 on the pancreatic islet function of diabetic rats.METHODS: The Wistar rats were injected intraperitoneally with low-dose(25mg/kg) of STZ after high fat diet for 14 weeks.The rats were divided randomly into three groups: the non-diabetic control group were fed conventional diet, the untreated diabetes group and the Exendin-4-treated diabetes group(the Ex-4 group) .diabetic Wistar rats were treated with Exendin-4 for 4 weeks in the study, which have also been compared with untreated diabetic rats and non-diabetic controls. Islet cell apoptosis,fasting blood glucose (FBG)and fasting insulin level (FINS) were detected and expressions of PDX-1, AKT, Bcl-2 and insulin in pancreatic islets were detected by immunohistochemical assay . We also studied alteration of the islet cell ultrastructure under Transmission Electron Microscope.RESULTS:(1) The untreated Diabetic rats had showed significant decreases of PDX-1, AKT and Bcl-2 expression compared with non-diabetic controls (P < 0.01). Compared with untreated diabetic rats and non-diabetic controls, FINS and expressions of PDX-1, AKT, Bcl-2 and insulin of Exendin-4-treated diabetic rats had been significantly increased (P<0.01).(2)The FBG of untreated diabetic rats increased as the study proceeded (P < 0.01). The FBG of Exendin-4-treated diabetic rats decreased as the study proceeded (P < 0.01).(3) FBG of untreated diabetic rats were significantly higher than non-diabetic...
