Objectives To evaluate the roles of heat shock protein 60(HSP60) and circulating anti-human HSP60 antibody in the initiation and progression of dilated cardiomyopathy(DCM) by detecting the expression of HSP60 in the DCM myocardial tissue and level of antibody against HSP60 in serum of DCM patients.Methods The expression levels of HSP60 in myocardial tissue in 15 DCM patients and 7 controls were detected by immunohistochemical staining and analyzed by computer image-pattern analysis system. The pathological changes of DCM myocardium tissue were observed by hematoxylin and eosin-staining. The serum anti-Hsp60 antibody levels in 44 DCM cases and 42 controls were measured by enzyme-linked immunosorbent assay(ELISA).Results (1) The hematoxylin and eosin-stained sections of myocardium samples showed that the hypertrophy of the ventricular myocytes and interstitial fibrosis were evident in DCM group compared to those in the control group. The expression of HSP60 increased significantly in the DCM hearts compared to the control hearts (P<0.05). Most of the HSP60 were observed in cytosol other than in nucleus or membrane. The average optical density(AOD)of HSP60 expression in the DCM group was 0.34±0.07, and it was noticeably increased as compared to that of the control group(0.25±0.05), P<0.05.(2) In comparison with the control group (8946.2±3976.6 ng/ml), there was an obvious increase in the serum anti-Hsp60 antibody levels of the DCM group (11695.8±2676.0 ng/ml), P<0.05.
