Objective To investigate the effect of reduced glutathione sodjum(GSH)on the expression of hypoxia-inducible factor-1α(HIF-1α) and nducible nitric oxide synthase(iNOS) in L-arginine(L-Arg)-induced acute pancreatitis(AP). And to discuss the relationship of HIF-1αmRNA and iNOS mRNA expression in AP. Methods Ninety-two Spraque-Dawley(SD) rats were divided into NS control group(group C, n=32) ,AP group(group A, n=32)and GSH group(group G, n=32) randomly. Group A and group G were induced by injecting intraperitoneally three times with 6% L-Arg (3×1.0mg/g) in an interval of 1 hour. Group C received saline alone, the dose and methods were same to group A. Group G was treated with 10% GSH(0.6mg/g)at 1 hour before the L-Arg induced. Rats were killed at 2nd, 6th, 12th and 24th hours after the last time of L-Arg injection. The expressions of HIF-1αmRNA and iNOS mRNA were assayed by reverse transcription polymerase chain reaction (RT-PCR). Pancreatic tissues were fixed in formalin. Histological characteristics were scored according to Rongione's standard.Results Compared to group A, serum levels of amylase in group G were significantly reduced when GSH was administered after induction of pancreatitis (P<0.05). Meanwhile, histological scores were significantly reduced at 12th and 24th hours. There were a few of HIF-1αmRNA and iNOS mRNA expressed in group C. In group A and G, the expressions of HIF-1αmRNA and iNOS mRNA were gradually increased with the time . Compared to group A ,the expressions of HIF-1αmRNA and iNOS mRNA in group G were significantly suppressed by GSH at 6th,12th,24th hours and there was a positive relationship between...
