| Ulcerative colitis (UC) is a kind of inflammatory bowel disease (IBD), whose etiology and pathogenesis are not very clear. Now, research findings considered that the pathogenesis should be related with heredity, immune, inflammatory and spirit ect, and the immunity was a very important aspect. Many researches are focusing on the immunity.Researches found that there were a lot of inflammation cells like lymphocyte, macrophage in UC patients' intestinal mucous membrane lamina propria. The humoral and cell immunity were activated when the UC occurred. There were also erythema nodosum, arthrophlogosis and iriditis self-immunologic disease when the UC took place. Adrenal cortical hormone could alleviate the disease symptoms. Some patients' serum could be found the antibody of anti-colon epithelial cell. All these had show that the immunity was an important role in UC.Because of the complexity of etiology and pathogenesis, the therapy of UC was also became difficult. The treatment drugs concluded hormone, sulphonamides, immunosuppressant ect. But the effects were not very good and there were also many side-effects. In recent years, traditional medicine has been used to treat UC. The traditional Chinese medicine regarded UC as "jiuxie, jiuli". Extro-xieqi, diet, spirit and deficiency of "piwei" made imbalance of "qixue" and "yingyang". In acute activing phase, the treat principal is expelling "xieqi". In chronic phase, the treat principal is supply "zhengqi" In this paper, DaHuang MuDan Decoction is an ancient recipe, which can clean heat and eliminate toxicant, treat "changyong". The experiment model in this paper is an acute phase model, so we choose DaHuang MuDan Decoction treat UC and study the immunity mechanism of treatment.Aims:1. At first, we set up the mice UC model and observed the bowel inflammation index, immune target and cell apoptosis. Researching the UC pathogenesis and the role of indexes in inflammation.2. After treated with DaHuang MuDan Decoction, we observed the indexes mentioned before, researched the pathogenesis of treatment with DaHuang MuDan Decoction.Methods:1. The establishment of UC mice model: we used 2,4,6 trinitrobenzene sulfonic acid and ethanol admixture made the female BALB/c mice create UC by rectum, detecting the mice weight, disease index, inflammation index and intestinal tract tissue pathological section.2. The treatment of UC: we treated the UC mice with DaHuang MuDan Decoction and detected the UC mice weight, disease index, inflammation index, observed the mice's thymus gland and spleen weight by detecting the thymus gland and spleen index variation, observed UC mice phagocyte, lymphocyte function variation by carbon clearance experiment, mice swelling of ear experiment and erythrolysin production experiment, observed UC mice cytokines TNF-ou IL-ip and IL-4 variation by ELISA and Luminex, and observed mice intestinal tract apoptosis variation by TUNEL end-labelling.Results:1. UC mice emerged pelage looseness, mind somasthenia, yellow loose stool in the second day of mould. In the 4th day, the loose stool symptom was relieved. In the procedure of mould, mice weight decreased and mean weight was lower than pro-mould. The disease index was 5.15±0.80. After one week, we executed and anatomized the mice, and found that the colon was adhesive with periphery tissue, enteric cavity distension, intestine wall was thickening, Mucous membrane edema, synathroisis and ulceration. Unaided eye pathologic histology score was 7.21±1.93 (total score is 10). Pathological section found part tissue was haematose and necrotic. Granulation tissue formed and cryptae was losing. Inflammatory cell infiltration and inflammation tissue score was 9.13±0.99 (total score is 10).2. After treated with DaHuang MuDan Decoction, mice disease index and inflammation index decreased significantly (2.45±0.80, 4.52±1.51). Pathological tissue score also decreased significantly (6.91±0.74) and inflammation symptoms improved remarkably. Thymus gland, spleen and carbon clearance index was increased significantly. Erythrocytolysin production, swelling of ear and serum TNF-cc > IL-ip decreasedremarkably. IL-4 level was increased but the result had no signification. Lymphocyte apoptosis was reduced in inflammation, but in this experiment, the result was not very good.Conclusions:1. TNBS-alcohol can induce mice form UC. The method is convenient and the disease symptoms are similar with patients" in clinic.2. DaHuang MuDan Decoction may mend UC inflammation symptoms and treat UC. It can improve inherent immunological functions, inhibit hyperfunction of cellular immune and pro-inflammatory cytokines production. DaHuang MuDan Decoction can mend UC inflammation symptoms by regulating UC mice immunological function.
