Objective To investigate the gene expression profile of CTGF (connective tissue growth factor ) in rat urinary bladder during growth in response to partial bladder outlet obstruction(PBOO).Material and Methods 36 healthy male Wistar rats were randomly divided into 3 group: BOO1 group, BOO2 group and the control group. In the former two groups, animal models of partial bladder outlet obstruction lasted 7 days and 14 days separately were made. In the control group, no ligature of the bladder and urinary was applied. At the end of the exercise, rat'bladders were harvested, and those samples'wet weight and Vmax were detected. Six of twelve rats in each group were selected randomly to be performed in the following experiments. The mRNA expression of CTGF was detected semiquantitatively with reverse transciption-polymerase chain reaction(RT-PCR). The localization of CTGF proteins was performed by immunohistochemical staining assay.Result Early bladder fibrosis plays an importante role in the increasing weight in response to PBOO. The mRNA expression of CTGF was significantly upregulated in obstructed bladder'tissue compared to those in control operation group (P<0.05). The mRNA expression of CTGF in BOO1 group was markedly upregulated than those in the BOO1 group (P<0.05). Immunostaining for CTGF was seen in all of BOO2 group and part of BOO1 group(3/6), which mainly is located within both lamina propria layers and surrounding the smooth muscle fascicles.Conclusions These data strongly indicate the involvement of CTGF in bladder wall remodeling as a result of the outlet obstruction. CTGF is involved in the progression of early bladder fibrosis following PBOO.
