Objective :to investigate expression of aquaporin in mouse colon after diarrhea different time and elucidate metabolism of aquafer in colon.Methods :40 KW mice were divided into control group and diarrhea groups randomly , diarrhea groups were given intragastric administration with 15% mannitol in which were examined at interval times 1h 8h 16h after diarrhea. Each mouse were dissected ascending,transverse,descending colon . Total RNA was purified from each colon ,and AQP8 mRNA expression was analyzed by fluorescent quantitative RT-polymerase chain reaction (FQ-RT-PCR). AQP8 protein expression was analyzed by immunohistochemistry.Result:1 immunohistochemistry revealed that AQP8 was expressed in all colon labeling of the surface epithelial cells and was localized in the apical part of the cell.2 FQ-RT-PCR analyses of AQP8 mRNA expression in rat colon, AQP8 was expressed in all colon and expression of descending is maximum in diarrhea and control groups. The AQP8 mRNA expression showed positive relationship with the degree and time of diarrhea. The AQP8 mRNA expression increased in diarrhea groups ,and in 8h diarrhea group is maximum,compared with others P<0.05.Conclusion:1 AQP8 was expressed in all rat colon .2 There are functional data suggesting that AQP8 is main role in colon water permeability . The AQP8 mRNA expression increasing in diarrhea groups is correlated with mannitol osmosis and stimulating.
