zinc finger protein 262 is essential for proper development of nerve system at embryonic stage, yet no literature concerning whether it is overexpressed in tissue of polycystic kidney disease (PKD) and its mechanism in the metanephric development has been published. We detected the expression and localization of ZNF262 mRNA in tissue of PKD and other associated cell line, we also investigated possible mechanism of ZNF262 in the metanephric development using RNA interference. Our studies reveal its overexpression in mesenchymal stem cell, tumor cell, embryonic kidney cell, PKD tissue and cell line, with significant correlation with PCNA. Moreover, its expression is gradually elevated with the development of PKD. Our data demonstrate that using RNA interference could facilitate the apoptosis of embryonic kidney cell line, up-regulating Bad, down-regulating PCNA, Bcl-2. all these suggest that ZNF262 may play a role in the pathogenesis of PKD and metanephric development, the possible mechanism of which is arresting apoptosis, promoting cell proliferation.
